主要研究領域

  • 第一項研究主題: 類風濕性關節炎 (rheumotoid arthritis, RA) 的致病機轉及可能治療方針, 主要內容包括:
  1. 針對具有致病能力之炎性介質,在關節滑液囊細胞(synovial fibroblast)及骨頭細胞製造過程的基因調控,包括signal transduction, transcriptional regulation, transcription factor/promoter interactions,或將某些基因予以over-expression, silencing, mutation後,對這些介質的製造有那些影響?
  2. In Vitro/ In Vivo correlation:在老鼠身上建立collagen-induced arthritis (CIA) 的disease model,依據”1”的結果,偵測這些可能抑制或促進RA的介質對CIA進展的影響,方法如下:
    a. 進行基因治療(gene therapy): 將特定介質的基因或該基因的small hairpin RNA(shRNA)置入lentivirus中,再以此濾過性毒注射老鼠,以對該基因做over-expression或 knock-out
    b. 我們最近也與其它實驗室合作,發展以奈米技術 (nano-technique)來包裝及傳輸基因,希望更提高gene therapy的效率。
    c. 針對特定介質, 與其它團隊合作來培育基因轉殖鼠(transgenic mice)。
    接著偵測a,b,c中CIA的病情發展,將以組織病理學配合影像分析,如:X光,電腦斷層去分析、偵測這些介質對CIA進展的影響,希望能由以上研究,發展RA的可能新治療策略。
  • 第二項研究主題: 我們也將建立其它在牙科醫療常見的disease model,如Chronic apical periodontitis, Biphosphonate-related osteonecrosis of the jaw (BRONJ), 並偵測這些在I-1所發現的介質在其中的表現。本實驗室合作的對象包括:
